Can I save over my existing sheet with an imported excel file?

Not that I am aware of unless you do via APIs. I've not tried that way before. 

Importing an excel file creates a NEW sheet.  So your existing data and formatting will disappear.

I'm having to do this on some existing excel sheets, so I am copying and pasting the data in as we have already created a template with conditionally formatting we want to retain.

You could have the vendor submit the data into the sheet vai a webform and then move their data around as needed.

What I did in the past was import the Excel sheet and then copy / paste the data into the Smartsheet sheet as Tim did.

I have played with the API and was able to update a sheet based on a csv exported from another system. This has to be customized, I think, because of sheet names, but is certainly do-able.

For me it made sense because that other system was not going away.

If this is a one-time task (and you are moving the system to Smartsheet as Tim and John suggest) then it likely isn't worth it.

If this is ongoing, then you should consider automation.
