I am trying to make a row highlight where the cell value does not contain L00 or X00 and highlight if anything else has been entered but cant seem to find how to do this. Any ideas

Would it work if you added the L00 and X00 as Not equal to and the Is not blank?

A possible workaround would be to add a so-called helper column with a formula checking a box if the above is true/false.

Not sure if this helps but at the bottom of the Set Condition screen there is an option for when condition is not met.
@ker9 just for the record, that option only exists on the "select from a list" menu and not the "define custom criteria" menu. There is no way to replicate "does not contain" solely in the Set Condition menu. You would need a dedicated helper column.
