I would like to count how many of my orders do not have PO number (i.e Po Number cell is blank)
Can you help with the formula to do this

Try this formula.
=COUNTIFS([PO Number]:[PO Number], ISBLANK(@cell))
Make sure your column title is PO Number. If it's different update my formula to match.
I would suggest adding in additional criteria for another column that is always populated. That way your formula doesn't include the blank rows at the bottom of the sheet.
If you have a date type column that will always have a date in it regardless of whether or not there is a PO Number, you could use something along the lines of...
=COUNTIFS([PO Number]:[PO Number], ISBLANK(@cell), [Date Column]:[Date Column], ISDATE(@cell))
