I am trying to set up a workflow for change cell when Complete is checked, status updates to 'Completed' but after setting this up saving and refreshing nothing happens when Complete is checked.
Glad we got it working! (except the delay)
If you continue to have a couple of minutes delay, I recommend reaching out to the Smartsheet Support Team.

Ensure that you've checked the Sheet Change Notification Settings box to Include my changes in sheet notifications in your Personal Settings (circle/profile in the lower-left corner).

I have just checked and this option is already selected. I tried unselecting and reselecting but to no luck.
