For example, I am asking users for their Supervisor's email address. From that entry in my form, I would like to send their Supervisor an approval email. Is that possible?

Yes. You will need to make sure that the column for the supervisor's email is type Contact List. From there, when you set up your automation, choose Notify Someone, and from the dropdown selection switch from "Send to Specific People" to "Send to a contact in a cell".
