Linking Job Done Column and Completion Column

Is there a ready function that I am totally missing to link the Job Done column to the Completion column? I just want to set it up such that when Job Done is ticked, duration will automatically updated with 100%, and vice versa - that when completion is marked as 100%, the job done will be ticked.

Try triggering it in the sheet itself. The "Run Now" feature ignores triggers and applies to every row based on the conditions within the automation. If there are no conditions in the automation, it will run on every single row.

You would need to set up change cell automations to have it working both ways.
