Can you create a project portal with a MS project file (MPP), and update the portal as you update the project file?

If I understand correctly, you want to attach a MPP project to your portal and make sure that it is always the current file.
If you are attaching a link to the MPP file using the Shortcut widget (connecting to Onedrive, Dropbox, etc) it will always be up to date since you are creating a link to the original file rather than uploading a specific version into Smartsheet.
