Is there a formula or reference for an assigned to column that would automatically be the sheet owner? I'm trying to setup an automatic reminder email to a sheet's owner two weeks before a milestone date.

You'll need to set up the Reminder for each sheet, and it won't change if the sheet owner changes.

Though that would be pretty neat.
Or if there was a way in your account to define a role (ie Head of HR) and if a new person takes that position, there would not be a lot of effort spent to update the emails.

Craig is correct, just set a sheet level reminder from the Alerts tab that sends the sheet owner (select the name manually) and email two weeks before the milestone date (select the column).
