So I set my project so that working days are Mon-Fri. However I want to make an exception on a specific Saturday, and make that a work day. Is that an option?

Sort of.
You can't have a task that starts earlier and carries over that particular Saturday (or Sunday), but you can allocate resources on that particular day.
You just need to add a 1 day (or less) task that starts and ends on the non-working day in questions. This also goes for "gotta work the weekend" tasks -- where you will need two 1 day tasks to allocate them properly.
